Roche Holding AG expanded its cardiometabolic development activity on September 24, 2026, when Atavistik Bio announced a strategic research collaboration with Roche covering novel small-molecule treatments for cardiovascular, renal and metabolic diseases. Atavistik Bio will conduct discovery and research activities, while Roche will handle further preclinical and clinical development, regulatory work and commercialization. Atavistik Bio said the agreement includes a USD 70 million upfront payment and potential research, development and commercial milestones of up to USD 1.9 billion, plus tiered royalties.
Cardiometabolic pipeline
The collaboration uses Atavistik Bio’s AMPS platform to identify allosteric small molecules against multiple targets. Roche said treating cardiovascular, renal and metabolic diseases remains a strategic priority, and its business development chief described the aim as developing oral small-molecule therapies from the partnership’s discovery work. The agreement therefore assigns the companies different stages of the program rather than announcing a completed product transaction. The release identified future net-sales royalties as part of the arrangement.
On September 22, 2026, Roche reported positive topline results from the randomized, double-blind, placebo-controlled Phase II CT-388-104 study of once-weekly enicepatide in 447 adults with type 2 diabetes and overweight or obesity. The study met both primary endpoints, measuring changes in HbA1c and body weight after 48 weeks. Roche’s release said the highest titrated dose was 24 milligrams.
At that dose, mean HbA1c fell 2.65% from a baseline of 8.1%, while mean weight loss reached 15.5% at week 48. The company also reported a 2.0% discontinuation rate because of adverse events in the enicepatide arms, compared with 0.0% in the placebo arm. Roche reported that common adverse events were predominantly mild-to-moderate gastrointestinal effects.
Kidney disease program
Roche also announced on September 23, 2026, positive prespecified interim results from the ongoing Phase III IMAgINATION study of investigational sefaxersen in adults with primary IgA nephropathy. The study met its primary endpoint, showing statistically significant and clinically meaningful improvement in proteinuria reduction versus placebo at 37 weeks, measured by the 24-hour urine protein-to-creatinine ratio. Roche said the study will continue to evaluate kidney function over two years.
Sefaxersen is an investigational once-monthly subcutaneous injection designed for self-administration. Roche described it as a liver-directed antisense oligonucleotide that inhibits production of complement factor B. The company said the safety and tolerability profile in the interim analysis was consistent with previously reported data and that no new safety signals were identified. The IMAgINATION study enrolled 459 people and is designed to assess treatment over 105 weeks. The company’s announcement said interim data will be presented at a medical meeting and shared with health authorities.
