Moderna's mRNA cancer vaccine cuts melanoma relapse risk by 49%, sending shares up 177% and triggering a wide range of analyst price targets from $25 to $170.

The numbers that emerged from Moderna's INTerpath-001 trial were stark enough to force a wholesale rethink on Wall Street. Over five years, the combination of the company's personalized mRNA cancer vaccine intismeran autogene with Merck's Keytruda cut the risk of relapse or death by 49 percent in roughly 1,137 patients with advanced melanoma. Both primary endpoints — relapse-free survival and metastasis-free time — were met, and a regulatory filing is expected within months.

What followed was one of the most dramatic repricings in recent biotech memory. Between August 18 and 19, the stock surged around 177 percent, lifting Moderna's market capitalization from roughly $25 billion to $60 billion. The shares have since cooled from that peak, but the debate over whether the valuation is justified has only intensified.

Wall Street's Great Recalibration

The analyst community has split into camps that are difficult to reconcile. Bank of America executed one of the most aggressive reversals of the cycle, moving Moderna from Underperform to Neutral while lifting its price target from $40 to $170 — a more than fourfold increase. Alec Stranahan, the analyst behind the call, had been among the skeptics until the data landed.

UBS followed with a target hike from $50 to $150, holding its Neutral rating. Barclays' Eliana Merle described the moment as a "watershed" for mRNA oncology, raising her target from $48 to $125 while keeping the stock at Equal Weight. Wolfe Research upgraded from Underperform to Peer Perform, projecting that the intismeran platform could eventually generate $9.2 billion in peak sales across four cancer indications. William Blair's Myles Minter went furthest on conviction, upgrading to Outperform and calling the Phase-3 result a "transformative catalyst" that validates the broader mRNA oncology platform.

Yet the consensus hides a much wider dispersion. JPMorgan sits at $77, while Wolfe Research's more cautious internal view lands at $25. The full range of analyst targets now spans from $25 to $170, with a mean of roughly $86.59. That gap is not a rounding error — it reflects fundamentally different views on whether one successful melanoma trial justifies a platform-wide re-rating.

Should investors sell immediately? Or is it worth buying Moderna?

The Technical Picture Tells Its Own Story

The stock closed Monday at €119.08, down 4.2 percent, as profit-taking set in after the historic run. The pullback extended to roughly 4 to 5 percent during intraday trading depending on the exchange, with other biotech names like Myriad Genetics also giving back 5.8 percent.

The technical indicators suggest the move was overextended. The relative strength index sits at 68.7, and the 30-day volatility has reached an extraordinary 517 percent on an annualized basis. From the 52-week low of $19.36 hit in November 2025, the shares have more than sextupled. At Monday's close, the stock sat about 20 percent below its 52-week high of $149.62, reached on August 19. On the most recent trading day, the stock added 11 percent to reach €131.78, narrowing that gap to roughly 12 percent.

The Fundamentals Beneath the Frenzy

The oncology breakthrough has also cast a long shadow over the rest of Moderna's news flow — some of it encouraging, some of it less so.

The company's quarterly results, released Thursday, showed a net loss per share of $1.97, beating the analyst consensus of $2.03, on revenue of $100 million. But the Norovirus vaccine candidate mRNA-1403 failed to meet early success criteria in a Phase-3 study — a setback that was largely drowned out by the oncology euphoria.

Regulatory progress continues on other fronts. The FDA approved mFLUSIVA (mRNA-1010), the first mRNA-based seasonal flu vaccine for adults aged 50 and older, which proved 27 percent more effective than standard shots in trials. However, the CDC's advisory committee ACIP has so far blocked a formal recommendation, leaving insurance coverage uncertain. Meanwhile, CEO Stéphane Bancel has warned of growing state-backed mRNA investment in China, even as the U.S. Department of Health and Human Services cut $500 million in mRNA funding.

Insider Moves and Valuation Questions

A recent SEC filing drew attention for what it revealed about insider activity. Bancel sold 499,246 shares in early August, worth approximately $28.7 million, to cover tax obligations and costs related to option exercises. He retained 252,469 shares from the exercise itself, bringing his direct holdings to 6.44 million shares. The sale occurred before the trial results were announced, suggesting routine portfolio management rather than strategic positioning. Over the past twelve months, insiders have sold nearly $48 million worth of stock.

The valuation debate remains the crux. GuruFocus calculates a price-to-sales ratio of 24.65 against a historical median of 8.38, and its fair-value model suggests the stock is overvalued by more than 470 percent. Analysts project annual revenue in the low single-digit billions by 2032, with optimistic estimates reaching $10 billion, based on treatment costs of around $300,000 per patient and gross margins between 50 and 80 percent.

The market is effectively pricing in a future that has not yet arrived — no approval, no commercial launch, no revenue from intismeran. Between the most bullish price target of $170 and the reality of an oncology pipeline still in its infancy lies a wide interpretive gap. For now, the stock remains a bet on whether a single Phase-3 success can translate into regulatory approval and commercial execution at scale. Until those milestones are reached, the volatility that has defined the past week is likely to persist.
