Moderna shares climbed 5.1% on Wednesday to EUR 175.00, extending a run that has been powered less by clinical breakthroughs than by the mechanical buying that accompanies an imminent index reshuffle. Index-tracking funds are being forced to build positions ahead of the biotech's entry into the Nasdaq-100, a move Nasdaq announced on Friday that takes effect before the opening bell on October 9. Moderna will displace media conglomerate Warner Bros. Discovery in the benchmark.

The advance stands apart from the broader vaccine sector, which offered no tailwind. Media reports indicate the gain was not part of a sector-wide move, and earlier rallies tied to brief concerns over disease cases at competitors such as Novavax had already faded the previous day.

Tempus Deal Builds Commercial Infrastructure

Alongside the index-driven flows, diagnostics firm Tempus announced Wednesday an expansion of its collaboration with Moderna and Merck to support an advanced oncology program. The multi-year agreement tasks Tempus with coordinating the collection and transfer of tumor tissue and blood samples for sequencing, and—subject to regulatory approvals—allowing the company to provide direct sequencing services. Financial terms were not disclosed.

The arrangement is aimed at supporting the potential commercialization of intismeran autogene, the personalized cancer therapy candidate Moderna is developing with Merck. Rapid processing of patients' genetic data is a critical component of both the clinical and logistical workflow for such individualized treatments, and the buildout of commercial infrastructure underscores how central tight diagnostic integration is to the platform.

Should investors sell immediately? Or is it worth buying Moderna?

A Split Analyst Picture

Not everyone is convinced the current valuation is justified. Geoff Meacham of Citi downgraded the stock from Neutral to Sell on September 30, even as he raised his price target to $80 from $60. Meacham grounded his negative call explicitly in what he views as a stretched valuation. Morgan Stanley took a milder stance on Wednesday, lifting its target to $95 from $89 while keeping an Equal-Weight rating. With two prominent houses setting targets at $80 and $95—well below current trading levels—the gap between analyst expectations and market pricing is widening.

The pipeline itself offers genuine reasons for optimism. Three abstracts on intismeran autogene, developed jointly with Merck, have been accepted for the ESMO 2026 congress, and Moderna has scheduled an investor webcast for October 24 at 1:00 p.m. EDT. The event coincides with the presentation of detailed melanoma study results at the ESMO meeting in Madrid. Yet an agreement covering logistics and sequencing is no guarantee of approval, let alone billion-dollar commercial revenue. The hope of an oncology breakthrough is real, but the market is already pricing it as though launch were imminent.

Leadership Transition and a Technical Overhang

Moderna also reshuffled its leadership this week. On Monday, Juan Andres stepped into the newly created role of Chief Operating Officer, tasked with steering the company's growing size and diversification, while Jerh Collins, the outgoing Chief Technical Operations and Quality Officer, retired. An SEC filing disclosed that Andres directly holds 125,000 shares as of Monday. Such transitions signal operational maturation but do little to resolve near-term valuation questions.

Adding to the technical picture, the stock has gained 3.9% since the Nasdaq-100 inclusion was announced roughly a week ago. Index funds must buy to mirror the benchmark, lifting the shares in the short term and layering mechanical demand over the fundamentals. At just 7.1% below its 52-week high, Moderna is trading at a level that leaves little room for disappointment.

The strategic alliance with Merck speaks to the technology's potential, yet the distance between incremental clinical milestones and the targets set by skeptical analysts remains wide. Once index-related inflows subside, the market is likely to return its attention to the underlying numbers—and the current strength looks more like a stretch of overheated confidence than a fundamental re-rating.
